Production of Grapevine Rootstock ¢¥5BB¢¥ Plants as Influenced by Bioreactor Culture Method and pH and EC of Nutrient Solution

Abstract
Shoot tips of grape rootstock ¢¥5BB¢¥ (selected at the Korean Fruit Research Institute) were cultured to obtain disease-free plantlets. The shoots were cut into single nodes (2 cm with a leaf), and cultured in bioreactors with three different culture method; continuous immersion (with or without air exchanges) and temporary immersion (with air exchanges). Shoot growth was greatly affected by air exchanges in bioreactor culture, plantlets were cut into single nodes (4 cm-long with a leaf), transferred to ex vitro and grown at different nutrient solution pHs and ECs. Compared to water supply, nutrient solution supply (1.6 to dS¡¤m^(-1)) induced 3-6 times greater shoot length, shoot dry weigh, root dry weigh, and total leaf area. Results also indicated that certain ions cannot be absorbed if the nutrient solution becomes too acid or alkaline. Plant growth was significantly different according to the pH of the nutrient solution: Shoot length, fresh and dry weigh, total leaf area, and CO©üuptake were greatest at pH6 but lowest at pH 4 and 8.
